Are post-mortem biochemical studies of human brain worthwhile?

et al., 1981), a value similar to that for the proteinase solubilized from myelin membranes. When the metalloproteinase activity from myelin membranes is purified to homogeneity it can be compared more accurately with meprin and endopeptidase 24.1 5. In addition, antibodies against this proteinase should permit immunoelectron microscopy to pinpoint its subcellular localization in the nervous system. We thank the Multiple Sclerosis Society for financial support, Nigel Groome (Oxford Polytechnic) for preparing mono-specific antibodies to MBP epitopes, Jeff Keen and John Findlay (University of Leeds) for N-terminal sequencing, and Nicola Woodroofe and Gillian Hayes (MS Society Laboratory) for immunohistochemistry.

Biochemical analysis of protein stability in human brain collected at different post-mortem intervals.
BACKGROUND & OBJECTIVES Rising prevalence of neurodegenerative disorders with a steady increase in aged-population necessitates studies of the human brain to understand their pathophysiology. As animal models are not available, medical centers have established "brain banks" to provide autopsy brain samples for such research. Frozen tissues must be of optimal quality to permit molecular and prot...
متن کاملTraumatic Brain Injury Has Not Prominent Effects on Cardiopulmonary Indices of Rat after 24 Hours: Hemodynamic, Histopathology, and Biochemical Evidence
Background: Accidents are the second reason for mortality and morbidity in Iran. Among them, brain injuries are the most important damage. Clarification of the effects of brain injuries on different body systems will help physicians to prioritize their treatment strategies. In this study, the effect of pure brain trauma on the cardiovascular system and lungs 24 hours post trauma was assessed. M...
متن کاملEffect of post-mortem delay on N-terminal huntingtin protein fragments in human control and Huntington disease brain lysates
Huntington disease is associated with elongation of a CAG repeat in the HTT gene that results in a mutant huntingtin protein. Several studies have implicated N-terminal huntingtin protein fragments in Huntington disease pathogenesis. Ideally, these fragments are studied in human brain tissue. However, the use of human brain tissue comes with certain unavoidable variables such as post mortem del...
متن کاملAltered glial marker expression in autistic post-mortem prefrontal cortex and cerebellum
BACKGROUND The cellular mechanism(s) underlying autism spectrum disorders (ASDs) are not completely understood, but ASDs are thought to ultimately result from disrupted synaptogenesis. However, studies have also shown that glial cell numbers and function are abnormal in post-mortem brain tissue from autistic patients. Direct assessment of glial cells in post-mortem human brain tissue is technic...
متن کاملA perspective on terra incognita: uncovering the neuroanatomy of the human subcortex
Recent exciting advancements in the field of in vivo neuroimaging allow for visualization of the living human brain with unprecedented anatomical detail. Large consortium studies will provide us with novel insights in the function and connectivity of the human brain. However, it is unlikely that the spatial resolution obtained using in vivo imaging will, in the near future, approximate the leve...
